Ascending and descending colon (AC and DC) and blood samples are obtained at baseline, and 26 and 52 weeks post randomization. Using flow cytometry, the following biomarkers are measured: RNA expression of TMPRSS2-ERG, a newly-identified and well-validated genomic determinant of cancer in both prostate and colorectal cancer; CD4+ and CD8+ T-cell counts; and soluble markers of inflammation, angiogenesis and immune response.
Metastatic colorectal cancer (mCRC) is typically treated with a combination of chemotherapy and targeted therapy. Randomized trials have demonstrated that patients with early stage disease (locally advanced and metastatic) can be treated with single agent (biologic or chemotherapy) agents. New treatment options are needed for patients with mCRC who fail treatment with irinotecan and a fluoropyrimidine-based chemotherapy backbone. This study will evaluate the clinical efficacy of two of these agents, aflibercept (VEGF) and bevacizumab (VEGF) in combination with chemotherapy versus chemotherapy alone in patients with mCRC.
The purpose of the study is to determine the safety and efficacy of the combination of aflibercept plus bevacizumab, and chemotherapy (5-FU/capecitabine or capecitabine/oxaliplatin) or capecitabine alone in patients with mCRC. The primary endpoint will be the duration of objective response, and the secondary endpoints will include progression free survival (PFS), overall survival (OS), time to progression (TTP), and time to treatment failure (TTF) which will be defined by the criteria of RANO (Response Assessment in Neuro-Oncology).

In developing the model, we considered the features that are typically expected in a learning portal and investigated the requirements and outcomes of applying this approach to a whole new learning environment: one designed to meet the needs of health professionals and of a cross-disciplinary group with expertise in musculoskeletal care. An online, multi-disciplinary portal with a staggered introduction of topics builds upon the existing educational resources which create a comfortable learning environment. It enables students to build their knowledge and research skills, as well as developing their communication and interpersonal skills.
